Study description:

The aim of the study was to evaluate penetration of NAD+ dissolved at 5% in distillate water (P1) vs. Intuisse Active NAD+ Liposomal Formulation (P2) on frozen human skin explants Perfex vivo, by infrared spectroscopy. Hyperspectral images using infrared spectroscopy (FTIR) are able to detect the active ingredient under the skin surface. By applying infrared radiation (IR), FTIR analysis measures the skin sample’s absorbance of infrared light at various wavelengths to determine the material’s molecular composition and structure. Analysis showed better penetration, in both epidermis and papillary dermis, on skin section treated with Intuisse Active NAD+ (P2) as compared to the non-liposomal NAD+ in water (P1).

In the images, the colour bar indicates that red is related to high values while blue is connected to low values.

Conclusions:

Analysis of NCLS (Non-negativity Constrained Classical Least Squares) score (% contribution of the active ingredient spectra within each spectrum in the hyperspectral images) shows that Intuisse Active NAD+ (P2) induces a significant increase by 30%** of the active ingredient Active NAD+ as compared to the non-liposomal NAD+ in water (P1).

Analysis at spectrum band 1030 cm-1

On day 1:

On the blank batch, a very slight signal on both stratum corneum and epidermis.

On the batch treated with NAD+ in water (P1), a slight signal of the band around 1030 cm-1 is observed in the stratum corneum and very slightly is observed in the epidermis.

On the batch treated with Intuisse Active NAD+ (P2), a fairly clear signal of the band around 1030 cm-1 is observed in the stratum corneum and a moderate signal is observed in the epidermis.

Analysis at spectrum band 2930 cm-1

On day 1:

On the blank batch, the ratio 2930/2960 cm-1 is weak in the epidermis and very weak in the papillary dermis.

On the batch treated with NAD+ in water (P1), the ratio 2930/2960 cm-1 is weak in the epidermis and very weak in the papillary dermis.

On the batch treated with Intuisse Active NAD+, the ratio 2930/2960 cm-1 is moderate in the epidermis and weak in the papillary dermis.

Decreases the number of non-functioning (senescent) cells

Study description:

The aim of the study was to examine the impact of Intuisse’s 5% liposomal formulation on the following three bio-markers using human living skin explants ex vivo.

Laminin-V is a basement membrane glycoprotein and a key component of the anchoring complex connecting keratinocytes to the underlying dermis. Laminin-V contributes to the resistance of the epidermis against frictional stress but also to basement membrane regeneration and repair of damaged skin.

Elastin is a protein of the connective tissue responsible for the elastic properties of the skin. The principal function of elastin is its ability to elastically extend and contract in repetitive motion when hydrated.

Elastin is the major component (90%) of the elastic fibers of the skin. UV exposure is able to damage the elastic network of the dermis, including elastin.

Collagen I is the main component of collagen fibers. Collagen I levels decrease not only during the ageing process, but also in chronically sun-exposed human skin.

Conclusion:

Laminin-V:

After 10 days of treatment and compared to the untreated condition, the product Intuisse (5% NAD+-Liposomal formulation) induces a significant increase of laminin-V along the dermal-epidermal junction, by 106%**. In comparison, retinoic acid in the culture medium at 0.1 μM for 9 days induced only 68% increase.

Elastin:

After 10 days of treatment and compared to the untreated condition, Intuisse (5% NAD+-Liposomal formulation) induces a significant increase of elastin fibers in the papillary dermis, by 35%** (comparable to retinoic acid).
